Abstract

The blood-brain barrier (BBB) is a highly selective permeability barrier that protects the brain from potentially harmful substances, while regulating the passage of nutrients and essential molecules. Recent advances in neuropharmacology have focused on overcoming this barrier, with natural products emerging as a promising class of compounds capable of crossing the BBB. This review explores the latest developments in the field, highlighting the mechanisms by which natural products can penetrate the BBB and their potential to treat a variety of neurological conditions, including Alzheimer’s disease, Parkinson’s disease, and stroke. We examine the role of bioactive molecules from plants, fungi, and marine organisms in facilitating BBB penetration, as well as their pharmacokinetic properties, stability, and therapeutic efficacy. The paper also discusses strategies for enhancing the bioavailability of these compounds through nanotechnology, drug delivery systems, and molecular modifications. By investigating the molecular and cellular mechanisms involved, this review sheds light on how natural products are opening new therapeutic avenues for CNS diseases, offering hope for the development of safe and effective brain-targeted therapies.
